Isoalantolactone reduces α-toxin production by Staphylococcus aureus

摘要:

为了考察异土木香内酯对金黄色葡萄球菌的抗菌活性,并研究其对金葡菌α-溶血素表达的影响。应用肉汤微量稀释法测定异土木香内酯对金葡菌标准株的最低抑菌浓度,采用溶血试验、Western-blot分析和荧光定量PCR分析测定异土木香内酯对金葡菌α-溶血素表达的影响。结果表明,异土木香内酯在2, 048 μg/ml仍然不能抑制金葡菌的生长,然而低浓度(1-8 μg/ml)的异土木香内酯能够剂量依赖性地降低金葡菌α-溶血素的表达。该研究提示异土木香内酯可作为以毒力因子为靶标的抗感染药物开发的先导结构。

Abstract:

In the study, we are aimed to investigate the antimicrobial activity of isoalantolactone against Staphylococcus aureus, and further assess the influence of isoalantolactone on the production of α-toxin by S. aureus. The minimal inhibitory concentrations (MICs) were determined by a broth micro-dilution method. The effects of isoalantolactone on α-toxin production were assessed by hemolysis, western blot, and real-time RT-PCR assays. The MICs of isoalantolactone against S. aureus strains were greater than 1,024 μg/ml. However, low concentrations of isoalantolactone could substaintially inhibit the expression of α-toxin by S. aureus. The data present here indicate that isoalantolactone may potentially be used as a lead compound for the development of anti-virulence drug.
