大叶藻组织培养中外植体消毒方法的研究

Study on the disinfection method for tissue culture of Zostera marina

摘要: 为探寻大叶藻组织培养中外植体的最佳消毒方法,采用75%乙醇、1.5%碘化钾、4%次氯酸钠和10%双氧水这4种消毒剂以及不同的处理时间组合,对大叶藻种子和幼苗进行消毒试验。试验结果,最有效的外植体消毒方法为:对大叶藻幼苗,先用无菌水冲洗3~4次,再以75%乙醇处理20 s,然后再用无菌水冲洗3~4次,可以达到较好的消毒效果。对大叶藻种子,先以75%乙醇处理10 min,再以1.5%碘化钾处理10 min,然后用无菌海水冲洗3~4次,这样消毒效果最佳,且不影响种子的发芽率。

Abstract: In order to establish the suitable disinfection method for tissue culture of Zoster marina. 75% ethanol, 1.5% potassium iodide, 4% sodium hypochlorite and 10% hydrogen peroxide as well as different disinfection time were used for seeds and seedlings sterilization treatment. The results indicated that for seedlings, sterile sea water rinsed for 3-4 times, 75% ethanol sterilized for 20 s, then sterile sea water rinsed for 3-4 times, which achieved good sterilizing effects. The efficient disinfection method for seeds was that, 75% ethanol rinsed for 10 min, 1.5% potassium iodide immersed for 10 min and then sterile sea water rinsed for 3-4 times, which showed the optimal disinfection effects and did not affect seed germination rate.
