珙桐的外植体的选材及组织快繁启动

Explant Selection and Micro Propagation Start of Davidia involucrata

摘要: 珙桐(Davidia involucrata)是一种濒危的孑遗植物,由于其花形优美,在园林绿化中具有很大的应用价值。但珙桐种子繁殖或扦插等繁殖方式都非常困难,无论从种质保护还是经济应用的需要,开展珙桐的快速繁殖组织培养技术研究都十分重要。本研究对珙桐组织培养的外植体选择、消毒液升汞的不同浓度和基本培养基进行了详细研究。结果表明,选择春季的鳞芽及半木质化的带芽枝条效果较好,75%的酒精消毒10 s 及0.1%升汞消毒10 min 基本能达到要求,而基本培养基以WPM培养基效果最好。研究结果构建了珙桐的以芽繁芽技术体系,但组培苗的生根需要进一步研究。

Abstract: Davidia involucrata is an endangered ancient relic species. As the flower shape of Davidia involucrata is graceful, it has great application value in gardening. But Davidia involucrata has obstacles in seed and shoot cutting propagation. Therefore, it is urgent to develop the technology of plant tissue culture for establishing a fast propagation system according to the needs of both germplasm conservation and economic application. In this research, explant selection, different concentrations of mercuric chloride disinfectant and basal culture media were explored. The results showed that spring bud and semi-lignified branches with bud were the best explants, and 75% alcohol disinfection for 10 s and followed by 0.1% HgCl2 disinfection for 10 min could meet the need, and the WPM basal media had the best effect. The results built the bud proliferation technique system, but the rooting of cultured seedlings need to be further studied.
