摘要: 甘露聚糖酶是一类水解甘露聚糖的关键酶,广泛存在于动植物和微生物中,细菌来源尤为广泛。为了更好地满足工业生产的需求,异源表达细菌来源的甘露聚糖酶基因,以获得性质优良的甘露聚糖酶。本文对甘露聚糖酶的来源和酶学性质进行了概述,归纳了细菌甘露聚糖酶的异源表达在提高酶活、扩大pH作用范围、改善安全性等方面的作用,包括对埃希氏大肠杆菌、枯草芽孢杆菌及毕赤酵母进行异源表达等。利用微生物资源拓宽甘露聚糖酶的应用领域,使用微生物技术提高产酶量满足工业需求,并为甘露聚糖酶定向进化和改造奠定基础。

Abstract: Mannanase is a key enzyme for hydrolyzing mannan, and is widely found in plants, animals and microorganisms, particularly in bacteria. In order to meet the needs of industrial production, heterologous expression of bacterial-derived mannanase gene is studied in order to obtain high-quality mannanase. In this paper, the origin and enzymatic properties of mannanase were reviewed, and the heterologous expression of bacterial mannanase was summarized concerning the aspects of improving the enzyme activity, expanding the pH range and improving the safety, including heterologous expression of Escherichia coli, Bacillus subtilis and Pichia pastoris. The use of microbial resources can broaden the application of mannanase, and the use of microbial technology can improve the amount of enzyme production to meet industrial needs, these lay a foundation for the determinate evolution and transformation of mannanase.
