中药复方宫净对奶牛子宫内膜上皮细胞抗炎作用研究

Effect of Chinese Herbal Compound Prescription Gong Jing on the Anti-inflammatory in the Bovine Endometrial Epithelial Cell

摘要: 为探讨中药复方的疗效和调控机制,本试验利用脂多糖建立奶牛子宫内膜上皮细胞炎性损伤模型,通过MTT法筛选出脂多糖最佳作用时间浓度时100 ?g/mL,12 h;中药复方宫净的最佳作用浓度为2 mg/mL。利用Real-time PCR技术检测对照组、模型组、中药组在3、6、12 h时细胞因子IL-1β、IL-6、IL-8 mRNA的表达变化。结果表明:与对照组相较,6、12 h模型组:IL-1β、IL-6、IL-8极显著升高(P<0.01);3 h:IL-1β、IL-8显著升高(P<0.05);中药组与模型组相较,3 h时IL-1β、IL-6显著下降(P<0.05);6 h时 IL-6、IL-8极显著下降(P<0.01),12 h时IL-8

Abstract: To learn the effect and mechanism of Chinese herbal compound prescription. In this study, LPS was used to establish endometrial epithelial cell model of cow inflammatory injury, 100 ?g/mL lipopolysaccharide was chosen to treat the epithelial cell for 12 h, which was screened by MTT assay. Chinese herbal compound prescription Gong Jing of 2 mg/mL was used to cure cow inflammatory injury. Then IL-1β, IL-6 and IL-8 mRNA was detected at 3h, 6 h and12 h respectively by Real-time-PCR. The results showed that compared with the control group, IL-1β, IL-6 and IL-8 was significantly increased at 6 h, 12 h of model group (P<0.01); IL-1β and IL-8 was significantly elevated at 3 h of model group (P<0.05). Compared with model group, Gong Jing made IL-1β, IL-6, IL-8 decrease significantly to 3h model group (P<0.05, P<0.01), while IL-6, IL-8 reduced notably to 6 h model group (P<0.01), and only IL-8 decreased to 12 h model group(P<0.05). In conclusion, LPS could induce IL-1β, IL-6, IL-8 mRNA expression, and traditional Chinese medicine had a certain anti-inflammatory effect by modulating the IL-1β, IL-6, IL-8 mRNA expression.
