龙眼叶片中CHI和CHS基因启动子序列的克隆与分析*

Cloning and sequence analysis of CHI and CHS gene promoter from longan (Dimocarpus longan) leaf

摘要:为探明龙眼叶片CHI和CHS基因在类黄酮物质合成过程中的作用,本研究应用染色体步移技术,克隆了642bp的 CHI启动子序列CHI-P和613bp的CHS启动子序列CHS-P。利用PlantCARE软件对克隆获得的启动子序列进行分析,结果表明CHI-P和CHS-P均含有核心启动子元件TATA-Box和CAAT-Box。Box I,ABRE,G–Box元件是CHI-P和CHS-P共有的顺式作用元件,这些元件的功能包括参与脱落酸响应(ABRE)与光反应,说明CHI和CHS的表达可能都受到脱落酸和光的调控。此外,CHI-P和CHS-P序列中含有的其他元件说明CHI和CHS的表达还与植物激素及外界环境的诱导密切相关。

Abstract:

Abstract: In order to study the role of CHI and CHS in flavonoids synthesis, a 642bp CHI promoter partial sequence and a 613bp CHS promoter partial sequence were cloned by genome walking PCR. The sequences were analyzed by PlantCARE software. The results showed that the core promoter elements TATA-Box and CAAT-Box were found in both sequences. The cis-acting elements including Box I, ABRE, G-Box were also found in both sequences. The functions of these elements were involved in the abscisic acid responsiveness and light responsiveness. It was indicated that the expression of CHI and CHS was controlled by abscisic and light. The results also found that the expression of CHI and CHS was related to plant hormones and environment.
