Abstract: The sugarcane tender leaves were taken as experiment materials, through made the improvement and the experiment to predecessors CTAB methods to reform a better extraction method of sugarcane genomic DNA. The final improved CTAB method (method Ⅲ) had the characteristics of costing low, operation simple, time-saving and so on. Ultraviolet absorbance spectrum analysis, agarose gel electrophoresis and PCR technique showed that DNA samples obtained by method Ⅲ had a good integrity and high purity, could be used in each kind of molecular markers directly.
