Phylogenetic Study on Cytochrome b in Cherry Valley Duck

摘要: 采用PCR技术对13个从国外引进的樱桃谷鸭的Cytb基因进行了克隆,测序结果基于Kimura 2-parameter模型进行遗传距离的计算,平均遗传距离0.004,通过DnaSP共检测出18个位点碱基存在变异,其中包括3个简约信息位点;可构成两种主要的单倍型,选取8个鸭品种并用构建了部分鸭科动物的系统发育树,发现樱桃谷鸭与棕颈鸭、太平洋黑鸭、麻斑鸭、野生黄嘴鸭归为一类,具有较高的亲缘性,利用简约信息位点可将8个鸭品种分为三个单倍型,因此本研究的结论可以用于鸭科动物的遗传标记筛选和群体的遗传多样性分析。

Abstract: The PCR technique was used to clone the Cytochrome b gene in thirteen individuals of Cherry Valley Duck breed. The genetic distances of thirteen sequences from Cherry Valley Duck were calculated based on the Kimura 2-parameter model, The overall average is 0.004, eighteen singleton variable sites were detected, 3 parsimony informative sites included. Two haplotypes were identified based on the three parsimony informative sites,with other eight duck breeds were selected from GenBank to construct phylogenetic tree, the Cherry Valley Duck and Anas superciliosa rogersi, Anas melleri, Anas luzonica, Anas undulate were ascribed into one group,The three parsimony informative sites can also classified the duck group into five haplotypes, the conclusions of our study can be used to screen genetical marker and analysis of duck group on genetic diversity.
