Applications of Plant Isolated Microspores and Microspore-derived Tissues in Genetic Transformation

摘要: 植物游离小孢子及其培养所获得的组织是植物基因工程中优良的受体,它们具有单倍体和较高胚胎发生能力的特性,以它们为受体构建遗传转化体系可快速获得纯合的转基因植株。概述游离小孢子及其培养所获得的胚状体、愈伤组织作为转化受体在根癌农杆菌介导法、基因枪轰击法、激光微孔穿刺法、显微注射法、电激法、PEG介导法等转化技术中的应用及存在的问题。

Abstract: Based on their characteristics of haploid and high embryogenetic capacity, plant isolated microspores and microspore-derived tissues are the good acceptors in plant genetic engineering, by which the homozygous transgenic plants could be obtained more quickly. The applications of plant isolated microspores, microspore-derived embryos and microspore-derived calli are reviewed and their existing problems are discussed in constructing genetic transformation system, including the detailed transformational technology of Agrobacterium mediation , particle bombardment, laser microbeam puncture, microinjection, electroporation, polyethylene glycol(PEG)-mediated.
