三种暖季型草坪草基因组DNA的提取方法

An improved method of extracting genomic DNA from Warm season Turfgrass

摘要:

采用改进CTAB法分别提取狗牙根(Cynodon dactylon)、结缕草(Zoysia spp.)、假俭草(Eremorchloa ophiuroides) 3种暖季型草坪草基因组DNA。得到的DNA经核酸蛋白质分析仪测得A260/A280值处于1.74~1.98之间、A260/A230集中处于1.74~1.99,经琼脂糖凝胶电泳检测各样品具有一条明亮主带并无拖尾现象。表明改进CTAB法提取得到的DNA完整性好,纯度高,适用于分子生物学的研究。

Abstract:

Genome DNA was extracted by the inproved method from leaves of Cynodon dactylon(L.) Pers,Zoysia spp,Eremorchloa ophiuroides.The detection performed on a nucleic acids and protein analyzer showed that A260/A280 were situated between1.74~1.98,A260/A230 were ranged from 1.74 to 1.99.The extracted products had a bright band and without tailing bands which could be observed through agar gel electrophoresis. These results showed that genomic DNA obtained by optimum methodWhich had a good integrity and high purity,and could be used in each kind of molecular biology research.
