摘要:

本研究在常规的CTAB法基础上,对其中某些步骤和所用药品进行改良,以寻求一种能简便快速地排除多酚类、蒽醌类和甾体类化合物等物质干扰的方法。应用该改良CTAB法所得鹧鸪茶总DNA鹧鸪茶总的DNA长度接近21kb,OD260/OD280在1.8左右。以此DNA为模板可获得条带清晰、重复性好的ISSR-PCR扩增结果。

Abstract:

Based on the routine CTAB method, some steps and reagents used to obtain DNA, were modified in this study to develop a simple and rapid method for removing the interference of some chemicals, such as polyphenols, anthraquinones, sterids and so on. The molecular weight of DNA, isolated by modified CTAB method, was nearly 21 kb, and the OD260/OD280 was about 1.8. Using the DNA isolated by modified CTAB method as the template in ISSR-PCR, the excellent amplification result, with distinct band and fine repeatability, could be obtained.
