枯草芽孢杆菌B-332菌株发酵条件的研究

Research on the Fermentation Conditions of Bacillus Subtillus B-332

摘要:

为了探明生防菌株枯草芽孢杆菌B-332的发酵条件及培养基配方,采用单因素和正交试验设计,通过摇瓶培养对枯草芽孢杆菌B-332菌株的发酵培养基和培养条件进行优化。结果表明,最适培养接种时间为18 h,优化后的培养基配方为:豆饼粉0.60 g/L、蔗糖0.25 g/L、硫酸铵0.07 g/L、柠檬酸三钠0.03 g/L、磷酸氢二钾0.003 g/L、硫酸镁0.005 g/L、硫酸亚铁0.0005 g/L;发酵条件为:温度30℃、摇床转速180 r/min、摇瓶装量80 mL/500 mL。在该综合条件下,发酵后的芽孢数为1.43×1011 cfu/mL,比初始条件的芽孢总数4.03×109 cfu/mL提高了34.48倍,为该菌株的工厂化打下了基础。

Abstract:

In order to ascertain the cultivated conditions and the optimal medial components of Bacillus subtilis B-332, the cultivated conditions for Bacillus subtilis B-332 were optimized by single factor and orthogonal experimental designs. The results showed that the optimal seed aged 18 h, and the optimal fermentation time was 72 h, the optimal medial components were composed of 0.60 g/L soybean oil meal, 0.25 g/L saccharose, 0.07 g/L ammonium sulfate, 0.03 g/L trisodium citrate, 0.003 g/L dipotassium phosphate, 0.005 g/L magnesium sulfate, 0.0005 g/L ferrisulphas. Under these conditions, numuber of spores was 1.43×1011 cfu/mL, and increased 34.48 times compared to initialization, and it formed the base for the factory.
