不同贮藏条件下孔雀石绿加标样品降解研究

Degradation of Malachite Green Standard Addition Samples Under Different Storage Conditions

摘要:

为筛选出验证药物品种的适宜储存条件、运输条件,以及最大限度消除外部因素对检测结果的影响,并保证结果的公正性,开展了水产品中孔雀石绿残留检测质量控制技术研究。本研究采用鳕鱼和鲤鱼2种不同基质,分析了孔雀石绿在3种浓度(1.5、5、50 μg/kg)和3种贮藏条件(常温25℃,冷藏4℃和冷冻-18℃)下随时间的降解规律。实验结果表明不同基质下,不同浓度的孔雀石绿因贮藏条件的不同,均有不同程度的降解,且降解速率各有不同。鳕鱼在3个浓度水平下的降解情况一致,均为高浓度的降解速率>中浓度的降解速率>低浓度的降解速率。鲤鱼在常温条件下,降解速率表现为高浓度的降解速率>中浓度的降解速率>低浓度的降解速率;在冷藏和冷冻条件下,降解速率均表现为中浓度的降解速率>低浓度的降解速率>高浓度的降解速率。说明在不同贮藏条件对孔雀石绿加标样品的影响不同,受酶作用的影响,短时间保存孔雀石绿加标样品可以常温保存,便于尽快进行检测;如样品不能尽快安排检测(时间超过48 h),样品须用冷冻方法进行保存,这样有利于保存样品的原有属性,避免对检测结果造成影响。

Abstract:

Quality control technology of malachite green residues in aquatic products were carried out to filter out the appropriate storage and transportation conditions for drug verification, with an aim to minimize the impacts of external factors on the tests results and to ensure the fairness of the outcome. Cod and carp were used as substrates to analyze the degradation of malachite green over time in 3 different concentrations(1.5, 5, 50 μg/kg)and 3 different storage conditions (normal room temperature 25℃, cold storage4℃ and freezing-18℃). The results showed that under different storage conditions, the degradation of malachite green with different substrates in varying concentrations varied and the degradation rate also varied. When cod was taken as the substrate, the degradation under three concentrations showed similar results: degradation under high concentration>degradation under medium concentration>degradation under low concentration. When common carp was taken as the substrate in normal room temperature, it was found that: degradation under high concentration>degradation under medium concentration>degradation under low concentration. However, under cold storage and freezing conditions, the result was different with degradation under medium concentration>degradation under high concentration>degradation under low concentration. It was concluded that different storage conditions had different effects on the malachite green standard addition samples. By the impact of enzyme, short-term preservation of malachite green standard addition samples could be kept at normal room temperature, which was also easy to conduct detection soon. If the samples couldn’t be arranged to be detected within 48 hours, the samples should be preserved by freezing methods, which was a good preservation of its original properties and also a good way to avoid impacts of this test results.
