超薄层等电聚焦电泳技术在杂交苦瓜种子纯度鉴定上的应用

Applying Ultrathin-layer Iso-electric Focusing Electrophoresis in Varietal Purity Testing of Hybrid Bitter Gourd

摘要:

为了发掘苦瓜(Momordica charantia L.)种子纯度室内快速鉴定方法,采用种子的水溶性蛋白超薄等电聚焦电泳与田间种植的方法对4个苦瓜杂交组合的F1种子纯度进行检验,并将结果进行一一比对,发现参试的4个组合的父母本均可找到特征蛋白质电泳谱带,而F1均含有父母本的特征蛋白质电泳谱带;超薄等电聚焦电泳技术鉴定的结果与田间种植鉴定的结果完全吻合,说明蛋白质超薄等电聚焦电泳技术可作为杂交苦瓜种子纯度检验的室内快速鉴定方法。

Abstract:

In order to develop a rapid and practical method for varietal purity testing in bitter gourd (Momordica charantia L.), four bitter hybrids were subjected to Ultrathin-layer isoelectric focusing electrophoresis (UTLIEF) and field grow-out test. Protein marker bands were found in all parent lines of the four hybrids, and the protein electrophoresis of these four F1 hybrids contained both male marker bands and female marker bands. The results of UTLIEF tests and grow-out test were compared, and the results were identical. This examination subjects that UTLIEF could be employed as a rapid and practical method for varietal purity testing in hybrid bitter gourd.
