Effect of Vascular Endothelial Growth Factor on in-vitro Maturation of Porcine Oocytes

摘要:

为了探讨适宜浓度的(Vascular Endothelial Growth Factor,VEGF)对猪卵母细胞体外成熟的影响,以提高猪卵母细胞的成熟效果进而促进胚胎的后期发育,以期为进一步改善猪卵母细胞体外成熟体系提供参考。本研究通过在猪卵母细胞体外成熟培养液中添加不同浓度的VEGF,成熟培养液(TCM199+BSA)中分别添加0 ng/mL VEGF对照组、5 ng/mL VEGF试验组Ⅰ、50 ng/mL VEGF试验组Ⅱ、500 ng/mL VEGF试验组Ⅲ的卵母细胞成熟培养后,经孤雌或体外受精和体外培养。结果表明:(1)试验组Ⅰ极显著地提高了猪卵母细胞的成熟率,试验组Ⅱ显著提高了猪卵母细胞的成熟率;(2)成熟的卵母细胞经孤雌激活和体外培养后,试验组Ⅰ极显著提高了孤雌胚胎的卵裂率和囊胚率。试验组Ⅱ显著提高了孤雌胚胎的卵裂率、极显著的提高了囊胚率;(3)成熟的猪卵母细胞经体外受精和体外培养后,试验组Ⅰ显著地提高了体外受精胚胎的卵裂率和囊胚率,试验组Ⅱ显著地提高了体外受精的卵裂率、极显著地提高了囊胚率。5 ng/mL或50 ng/mL的VEGF,有利于促进猪卵母细胞的体外成熟和早期胚胎的发育。

Abstract:

In this study, by adding different concentration of Vascular Endothelial Growth Factor (VEGF) on in-vitro maturation of porcine occytes, will investigate the suitable concentration of VEGF for the porcine oocytes and therefore improve its embryo maturity in the late stage, which would be the reference for the further improvement of the maturity of porcine oocytes. The different concentrations of VEGF were added into the maturation medium (TCM199+BSA), they were treated as following: the Control group: the porcine oocytes were cultured in the maturation medium without VEGF, GroupⅠwith 5 ng/mL VEGF, Group II with 50 ng/mL VEGF, Group III with 500 ng/mL VEGF respectively. Then, these oocytes were treated by parthenogenesis or in vitro fertilization and vitro culture respectively. The results were shown that: (1) the maturation rate of porcine oocytes in GroupⅠwas significantly higher than that of the control one, and the appropriate maturation rate in GroupⅡ was much higher than that of the control one; (2) The cleavage rate and blastocyst rate of their parthenogenetic embryos in Group I were significantly higher than that of the control one, in addition, this cleavage rate in Group II was much higher than that of the control one, as well as blastocyst rate was significantly higher than that of the control one; (3) While in their in vitro fertilization and in vitro culture, The cleavage rate and blastocyst rate of in-vitro fertilized embryos in Group I were all much higher than that of the control one, in addition, The cleavage rate of in-vitro fertilized embryos in Group II was all much higher than that of the control one, and the blastocyst rate was significantly higher than that of the control one. Collectively, it could be concluded that the concentration of 5 ng/mL or 50 ng/mL VEGF be beneficial to both in vitro maturation of porcine oocytes and their appropriate in vitro development.
