AtFT基因植物表达载体的构建研究

Construction of Plant Expression Vector of AtFT Gene

摘要: FT是从拟南芥克隆得到的诱导植物开花调控基因。利用FT基因表达特点,克隆拟南芥FT基因片段,重组构建了以CaMV35S为启动子、GUS为报告基因的植物表达载体pCAMBIA2301-FT与pCAMBIA2300-FT:GUS,并采用农杆菌介导法转化橡胶树体细胞胚,通过GUS组织化学染色观察橡胶树体细胞胚瞬时表达情况,结果验证了重组构建的2个载体的有效性,为进一步研究FT基因在橡胶树中的功能及应用奠定基础。

Abstract: FT was an important gene involved in controlling plant flowering time from Arabidopsis thaliana. This research used FT gene expression characteristics, through cloning FT gene from Arabidopsis thaliana, plant expression vector pCAMBIA2301-FT and pCAMBIA2300-FT:GUS which had a CaMV 35S promoter and GUS reporter gene, were successfully constructed. The somatic embryos in Hevea brasiliensia were transferred by agrobacterium-mediated transformation, and the instantaneous expression of somatic embryos was observed using GUS histochemical staining. The result showed the two plants expression vectors were effective. It laid the foundation for further studying the function and application of FT genes in Hevea brasiliensia.
