应用正交设计法优选台湾金线莲快繁培养基

Optimization of the Rapid Propagation Medium for Anoectochilus formosanus by Orthogonal Design

摘要: 以台湾金线莲试管苗顶芽为外植体,研究基本培养基、6-BA、NAA、蔗糖4个因素对不定芽增殖培养与生根培养的影响,应用L9(34)正交试验设计优选出台湾金线莲快繁培养基。结果表明,4个因素对不定芽增殖与生根培养均有极显著影响。对不定芽增殖培养的影响为:6-BA>蔗糖>NAA>基本培养基。对生根培养的影响为:蔗糖>6-BA>基本培养基>NAA。最适宜的增殖培养基为:MS+6-BA 3.0 mg/L+ NAA 0.3 mg/L+蔗糖20 g/L。最适宜的生根培养基为:1/2MS+6-BA 1.0 mg/L+NAA 0.6 mg/L+蔗糖40 g/L。

Abstract: It was studied on four factors of basic medium, 6-BA, NAA and sucrose on adventitious buds proliferation culture and rooting culture to optimize the rapid propagation medium of Anoectochilus formosanus by orthogonal design L9(34), taking the apical buds of test-tube seeding as the explants. The results showed that: four factors had a very significant impact on the adventitious buds proliferation culture and rooting culture. The effect for proliferation culture: 6-BA> sucrose> NAA> the basic medium, the effect for rooting culture: sucrose> 6-BA> the basic medium> NAA. The optimizing medium for proliferation culture was MS+6-BA 3.0 mg/L+NAA 0.3 mg/L+ sucrose 20 g/L. The optimizing medium for rooting culture was 1/2MS+ 6-BA 1.0 mg/L+ NAA 0.6 mg/L+ sucrose 40 g/L.
