茶梅花瓣总RNA提取方法的比较和分析

Comparison and analysis of methods of extracting total RNA from petals of Camellia sasanqua

摘要: 本研究分别利用CTAB多级沉淀法、改良的异硫氰酸胍法、改良的热硼酸法、Trizol法和EASY spin植物RNA提取试剂盒法等五种方法提取茶梅花瓣总RNA,并进行了对比分析,发现EASY spin植物RNA提取试剂盒法是最为简单、快速、高效的方法,CTAB多级沉淀法次之。本研究为今后有效开展茶梅分子生物学研究提供技术便利,并为其它花卉植物RNA的提取提供必要的参考。

Abstract: The total RNA from petals of Camellia sasanqua was extracted by the methods of CTAB-many-level-precipitation method, improved isothiocyanate method, improved hot-borate method, Trizol method and EASY spin plant RNA extraction Kit method, respectively, and the five methods were contrasted and analyzed. The results showed that the method of EASY spin plant RNA extraction Kit was the simplest, the quickest and high efficient, and CTAB-many-level-precipitation method took the second place. The study provided convenient and effective technical supports for the research on molecular biology of Camellia sasanqua, and also provided necessary reference for the RNA extraction of other flowers.
