台湾桤木外植体褐化机理初探

A Preliminary Study on browning mechanism of explants of Alnus formosana

摘要: 对台湾桤木组培褐变中的几个相关酶活性和总酚含量变化的影响进行研究,为台湾桤木组织培养技术做前提。以台湾桤木的腋芽或顶芽为外植体,研究Vc浓度、培养基pH值和外植体采集时间对褐化的影响。总酚含量、多酚氧化酶(PPO)、过氧化物酶(POD)与外植体褐化相关。其中添加0.1 g/L Vc的培养基、pH为(5.0~5.8)的培养基、上午10:00采摘外植体都能有效抑制褐化。通过不同的处理抑制总酚含量及相关酶的活性,从而减轻台湾桤木外植体的褐化。

Abstract: The aims were to study the related enzyme activities and the impacts of the change of the total phenolic, on the premise of the technology study of the tissue culture for Alnus formosanas. The axillary bud or crown of the Alnus formosana were used as explant, to study the concentration of Vc, medium PH value and the influence of the explants collection time to browning. Total phenolic, PPO and POD were all related to the browning of explants, among which added 0.1 g/L Vc medium or pH 5.0-5.8 medium or collect explants at 10:00, all these can effectively restrain browning. By processing the different total phenol content and the activity of relates enzymes, so as to reduce the browning of Alnus formosana.
