醋酸钙不动杆菌PAHs降解酶的超声波提取优化

Optimization of Ultrasonic disruption for PAHs-depredating enzymes from Actnetobacter calcoaceticus

摘要: 为了提取醋酸钙不动杆菌 (Actnetobacter calcoaceticus)的PAHs降解酶,本文利用超声波破碎醋酸钙不动杆菌细胞,对多环芳烃(PAHs)降解酶进行优化提取。在对超声输出功率、工作\间隔时间、菌体稀释倍数以及总超声时间进行单因素实验的基础上,利用Box-Behnke中心组合实验设计,响应面法优化得到多环芳烃降解酶的最佳提取工艺为:工作\间隔时间为2s\2s、超声功率320W、总超声时间17 min、稀释12.5倍,在此条件下提取的酶活力为95.55U/g。

Abstract: Ultrasonic wave was used in this paper to disrupt the cell of Actnetobacter calcoaceticus WG-6 for PAHs-depredating enzymes. The effect of ultrasound output power, duration of radiation each time, total irradiation time and the cell dilution multiple on the activity of PAHs depredating-enzymes were investigated through the single-factor experiment. The optimum conditions of PAHs depredating-enzymes extraction were determined through box-benhnken center-united design and response surface method were duration of radiation each time 2s (with a 2s interval), ultrasonic power 320W, total irradiation time 17 min, cell dilution multiple 12.5. The PAHs-depredating enzymes activity was 95.55U/g under this condition.
