Simultaneous Determination of Hederacoside C and α-Hederin in Hedera nepalensis var. sinensis by High Performance Liquid Chromatography-Evaporative Light-Scattering Detector

摘要: 为了提高常春藤皂苷检测的灵敏度,以中华常春藤为试验材料,采用HPLC-ELSD对中华常春藤中常春藤苷C和α-长春藤皂苷的分析方法进行了研究。采用的色谱条件为:Welchrom C18 (250 mm×4.6 mm,5 μm)色谱柱;水-乙腈溶液为流动相,梯度洗脱;检测器为蒸发光散射检测器;流速1.0 mL/min;柱温30℃。常春藤苷C的进样量在0.19188~2.3985 μg范围内峰面积的对数与进样量的对数之间线性关系良好;α-常春藤皂苷的进样量在0.15615~4.6844 μg范围内峰面积的对数与进样量的对数之间线性关系良好。该方法灵敏度高,具有良好的稳定性和准确度。

Abstract: In order to improve the detection sensitivity of saponins, a kind of high performance liquid chromatography(HPLC) coupled with evaporative light scattering detector(ELSD) was established for the determination of hederacoside C and α-hederin in Hedera nepalensis var. sinensis. A welchrom column (250 mm×4.6 mm, 5 μm) was used with the mobile phase of water and acetonitrile solution. The flow rate was at 1.0 mL/min. This method showed good linearity for both hederacoside C (ranging from 0.19188 to 2.3985 μg) and α-hederin (ranging from 0.15615 to 4.6844 μg). The results demonstrated that this method was precise, accurate and can be used for the determination of hederacoside C and α-hederin .
