PPARγ基因在荷斯坦公牛不同组织部位表达差异性研究

Differential Expression of PPARγ Gene in Different Tissues of Holstein Bulls

摘要: 为研究PPARγ基因在荷斯坦公牛组织及月龄间的相对表达差异性。运用实时荧光定量PCR法检测了PPARγ基因在15和17月龄荷斯坦公牛脂肪组织(背部脂肪、腰部脂肪、肾周脂肪、肠系膜脂肪)和肌肉组织(背最长肌和半腱肌)间的相对表达差异性。结果表明:PPARγ基因的表达在荷斯坦公牛不同月龄间没有发现差异性,而组织间差异表达分析表明:肠系膜脂肪、肾周脂肪和腰部脂肪的相对表达量显著高于背最长肌和半腱肌(P<0.05);肠系膜脂肪和肾周脂肪相对表达量显著高于背部脂肪(P<0.05)。由此可见,PPARγ基因在荷斯坦公牛不同部位脂肪组织和肌肉组织中均有表达且部位不同表达不同,存在部位差异性。

Abstract: The objective of this study was to compare the differential expression of the peroxisome proliferators-activated receptor-γ(PPARγ) gene in different tissues in two ages of Holstein bulls. Real-time fluorescent quantitative PCR was used to determine the relative expression of the PPARγ gene using RNA isolated from different adipose tissues (subcutaneous fat over the rib, subcutaneous fat over the sirloin, kidney fat, and mesenteric fat) and muscle tissues (longissimus dorsi and semitendinosus) in two ages (15 and 17 mo) of Holstein bulls. The results showed that, expression of PPARγ in mesenteric fat, kidney fat and subcutaneous sirloin fat was greater (P<0.05) than the longissimus dorsi and semitendinosus. And it was also greater (P<0.05) in mesenteric fat and kidney fat than the subcutaneous rib fat. But it had no difference between 15 and 17 mo in the same tissue. The expression of the PPARγ gene in tissues of Holstein bulls differed among sources of tissue. These results suggest that evaluation of gene expression of PPARγ gene should consider the sources of the tissue.
