人参果‘长丽’叶片再生体系的建立

Establishment of Leaf Regeneration System in Ginseng Fruit‘changli’

摘要: 为了建立人参果遗传转化再生体系,以‘长丽’品种试管苗叶片为材料,研究了培养基种类、pH对人参果试管苗生长和激素组合对人参果叶片不定芽再生的影响。结果表明:培养基pH 5.7~6.0时,MS培养基有利于人参果试管苗的生长,培养24天时平均株高达8.5 cm;GS培养基有利于试管苗生根,培养24天试管苗的生根量达到13.8个/株。6-BA与TDZ配合使用,可显著提高叶片不定芽的再分化能力,在MS+6-BA 2.0 mg/L+TDZ 0.5 mg/L培养基上叶片的再分化率达到83.3%,再分化系数达8.5。本试验研究发现人参果叶片不定芽再生的适宜培养基为MS+6-BA 2.0 mg/L+TDZ 0.5 mg/L。

Abstract: In order to establish the genetic transformation regeneration system of ginseng fruit, taking the seedlings leaves of‘Chang Li’as test material, the effects of types of culture medium, pH and hormones on the adventitious buds subculture proliferation of ginseng fruit were studied. The results indicated that MS medium was favorable to the tube seedling of ginseng fruit and the average plant height was 8.5 cm cultivated for 24 d. GS medium was the optimal condition for the root generation of the tube seedlings, and the number of root generation could reach 13.8 per shoot. 6- BA blended with TDZ could improve the generation rate of adventitious buds. The regeneration ratio of leaves was 83.3% on the MS+6-BA 2.0 mg/L+TDZ 0.5 mg/L medium, the regeneration coefficient reached 8.5. Therefore, MS + 6- BA 2.0 mg/L + TDZ 0.5 mg/L was the optimal medium and in favor of the adventitious shoot regeneration of leaves.
