油松不定根诱导研究

Induction of Adventitious Root in Pinus tabulaeformis

摘要: 为建立诱导油松不定根高频发生的实验系统,研究以组培不定芽和实生苗下胚轴插穗为材料,进行适合油松不定根诱导材料的筛选。在此基础上,以下胚轴为材料,研究子叶、苗龄、不同浓度的植物生长调节物质(IBA、BA)组合以及预处理对其生根的影响。结果表明,油松下胚轴插穗比组培不定芽具有更高的不定根发生潜力;以2周龄实生苗的下胚轴为材料,在1/2 DCR+ 20 g/L蔗糖+ 6 g/L琼脂+ 4 mg/L IBA、pH 5.8的培养基上预培养15天,然后转到去除IBA的相同成分的培养基上继续培养,可以获得较好的生根效果,此条件下生根率最高可达96.67%,这为进一步研究油松不定根发生的相关机理提供了可行的实验体系。

Abstract: To establish a model system with high frequency adventitious rooting in Pinus tabulaeformis, adventitious buds and hypocotyl cuttings from this species were used to induce adventitious roots. Then, effect of cotyledon, age of seedlings, plant growth regulators and treatment methods on the induction of adventitious roots from hypocotyls were studied. The results showed that hypocotyl cuttings have more stronger potential to induce adventitious root than adventitiou buds. And suitable medium was 1/2 DCR medium containing 20 g/L sucrose and 6 g/L agar with pH 5.8. When cultured the hypocotyl cuttings from 2-week old seedlings on this medium with 4 mg/L IBA for 15 days, then turned the culture to the same medium without IBA, the frenquency of adventitious rooting high to 96.67% was obtained. This provided a feasible experiment system for the further research on mechanism about adventitious root formation in Pinus tabulaeformis.
