正交设计优化PEG引发关苍术种子萌发的最优处理方案

摘要: 为提高关苍术种子发芽率,以不同PEG浓度、浸泡时间、浸泡温度及分子量为试验因素,采用正交设计筛选引发关苍术种子萌发的最优处理方案。结果表明:4种处理因素对关苍术种子萌发的影响程度依次为,处理温度、处理浓度、PEG分子量、处理时间;基于正交设计的关苍术种子PEG活性化处理方案中最优组合是A1B1C2D1,即关苍术种子在15℃条件下,浸泡在浓度为10%的PEG-4000溶液12 h时后,其发芽率可显著提高,且种子内部的SOD、POD和CAT酶的活性增强。

Abstract: The Atractylodes japonica Koidz ex Kitam seeds were soaked in different concentrations of PEG with different soaking time and molecular weight using orthogonal designs to select the best treatment scheme of PEG-induced germination percentage in order to improve the germination percentage.The results showed that in the four treatment factors which were treatment temperature,treatment concentration,molecular weight of PEG and treatment time decreased in turn on the effect of seed soaking in PEG on germination percentage and the best group of the treatment scheme was A1B1C2D1 that is,the germination percentage of seeds has improved prominently which were treated with PEG in concentration of 10% at 15℃ for 12h,and the activity of superoxide dismutase(SOD), the peroxidase(POD)and catalase(CAT) has increased.
