生防细菌SY286发酵条件优化

Optimizing Fermentation Condition for SY286 Strain

摘要: 为探讨生防细菌SY286 菌株液体发酵条件,提高其活性次级代谢产物的产量,以菌体生物量和发酵液抑制葡萄霜霉病菌(Plasmopara viticola)活性为指标,采用单因子试验和正交试验对菌株的最适发酵培养基成分及发酵条件进行优化。结果表明,菌株SY286 最适发酵培养基为葡萄糖10 g/L、牛肉膏7.5 g/L、氯化钠2.5 g/L、硝酸钾5 g/L;最佳发酵培养条件为培养温度27℃、培养时间72 h、初始pH 7.0、250 mL三角瓶装液量90 mL、接种量体积分数5%、摇床转速150 r/min。在最佳发酵培养基和培养条件下,菌株SY286发酵液抑菌率达到99.19%,抑菌能力提高6.98%。

Abstract: In order to investigate best condition for liquid fermentation of strain SY286 and increase its antagonistic substance productivity against Plasmopara viticola, the single factor experiments and orthogonal experiments were adopted to optimize the liquid fermentation medium and fermentation condition. The result showed that the optimum medium was glucose 10 g/L, beef extract 7.5 g/L, NaCl 2.5 g/L, KNO3 5 g/L. The optimal fermentation conditions were culture temperature 27℃, fermentation time 72 h, initial pH 7.0, medium capacity 90 mL/250 mL, inoculation rate 6%, and rotate speed 150 r/min. The inhibitory activity of strain SY286 fermentation solution increased by 6.98% under the optimum fermentation medium and culture conditions, it was up to 99.19%.
