摘要:

为分析嵌合猪圆环病毒(PCV1-2)对仔猪的致病性与免疫原性,检测了健康普通仔猪接种PCV1-2后的临床症状、病理变化、血清与组织中的病毒含量及血清学变化。结果显示,仔猪接种PCV1-2后增重和饲料转化效率未受影响,未出现可见的临床症状和大体病理变化,仅部分出现轻微的组织病理变化。血清中猪圆环病毒2型(PCV2)ORF2蛋白抗体在接种后第14天出现阳转,第28天时抗体阳性率达到100%,表明PCV1-2诱发仔猪快速产生了针对PCV2 ORF2蛋白抗原的特异性抗体反应。荧光定量PCR结果显示,血清中PCV1-2 DNA含量在接种后逐渐上升,第28天时达到最高,之后迅速下降;第35天宰杀时组织中病毒含量以肺门淋巴结中最高,心脏中最低;而常规PCR难以检出血清和组织中的PCV1-2 DNA,表明PCV1-2接种后只引起仔猪的轻微感染。研究数据表明,PCV1-2对仔猪具有良好的免疫原性,其致病性明显减弱。

Abstract:

To explore the pathogenicity and immunogenicity of chimeric porcine circovirus (PCV1-2) to piglets, several healthy conventional piglets were inoculated with PCV1-2, the clinical evaluation, gross pathology and histopathology, virus load in sera and tissues, and serology were determined. The results showed that there were no significant differences in the average gains and feed conversion efficiency of piglets between the PCV1-2-inoculation and the control, and there were not visible clinical signs and gross lesions in PCV1-2-inoculated piglets, although slight microscopic lesions were observed. The antibodies against porcine circovirus type 2 (PCV2) ORF2 protein in sera began to converse to positive on 14 days post inoculation (DPI), and were 100% positive on 28 DPI, which revealed that the PCV1-2 induced a quick specific antibody response to the PCV2 ORF2 protein antigen in piglets. The results of real-time PCR showed that the DNA loads of PCV1-2 in sera increased gradually and peaked on 28 DPI, declined quickly thereafter. The DNA load of PCV1-2 in tissues was the highest in lung lymph node and lowest in heart on 35 DPI, when the piglets were slaughtered. However, it was difficult to detect the PCV1-2 DNA in sera and tissues with conventional PCR, which suggested that the inoculation with PCV1-2 could only cause a slight infection of piglets. These data indicated that the chimeric PCV1-2 was with good immunogenicity to piglets but was attenuated obviously.
