刀豆氨酸的提取及测定方法的优化

摘要:

研究刀豆氨酸的提取及2,4-二硝基氟苯与刀豆氨酸在pH 9.5硼砂缓冲溶液中反应的最佳条件,进而优化测定刀豆氨酸的方法。运用分光光度法测定2,4-二硝基氟苯与刀豆氨酸的络合物在571 nm处的吸光值。结果表明,刀豆氨酸水提取法效果最好,DNFB-乙腈的用量为30 μL时,刀豆氨酸与DNFB反应的络合物在571 nm处有最大吸收值。在30 min内稳定以后该络合物的OD值随着时间的增长而增加,对应线性回归方程为y=0.0083x-0.052,相关系数R2=0.9986。

Abstract:

The extraction of L-canavanine and the best reaction conditions of L-canavanine and 2,4-dinitrofluorobenzene in pH 9.5 borax buffer were studied in this paper, which can optimization the method of determination. The OD value of L-canavanine and 2,4-dinitrofluorobenzene’s complex in 571 nm was determined by spectrophotometry. The results showed that the best extraction method of L-canavanine was water extraction. When the volume of DNFB-acetonitrile was 30 μL, the complex reaction absorption value of L-canavanine and DNFB in 571 nm was maximum. The OD value of the complex was increasing with the extension of time after stability within 30 minutes. Corresponding linear regression equation was y=0.0083x-0.052; Correlation coefficient was R2=0.9986.
