东北梅花鹿血液SOD纯化及部分性质的研究

Purification and Properties of SOD in Different Physiological Period of Sika Deer Blood

摘要:

为了研究生茸期梅花鹿血液中SOD纯化技术及其部分性质,为开发鹿血抗衰老资源奠定理论基础。用氯仿乙醇除血红蛋白、热变性、丙酮沉淀、DEAE层析等技术对生茸期鹿血SOD进行纯化;用SDS-聚丙烯酰胺凝胶电泳测定其亚基分子量。结果表明:SOD的初始比活力为12.84 U/mg,经纯化后得到SOD 0.16 mg/mL,最终比活力4122.74 U/mg,纯化倍数为321.2倍,回收率66.38%;鹿血SOD含有2个亚基,其相对分子量为17.32 KD和16.28 KD。结果提示:鹿血SOD在60℃,0.002 mmol/L的Cu2+浓度下SOD活力最高,此时所得SOD纯化效果较好。

Abstract:

Purification SOD in the blood of Sika Deer with in antler developing period and study on properties of SOD, were studied for developing anti-aging resources of deer blood and laid a theoretical basis. SOD has been purified from Sika Deer blood by chloroform-ethanol fractionation, thermal denaturalization, acetone precipitation, DEAE-32 chromatography and the SDS-PAGE was used to measure its subunit molecular weight. The results showed that SOD initial specific activity was 12.84 U/mg and 0.16 mg/mL-purified SOD was achieved. Finally, specific activity was 4122.74 U/mg ,and 321.2 times purification rate and 66.38 percent recovery rate were achieved. The relative molecular weight of subunit for SOD was 17.32 KD and 16.28 KD. The results demonstrated that the 60℃ and 0.002 mmol/L Cu2+ was the best condition for SOD purification.
