麦穗鱼不同组织DNA含量及细胞周期的分析研究

Analysis of DNA Contents and Cell Cycle of Different Tissues of Pseudorasbora parva

摘要:

为了解鱼类各组织DNA含量和细胞周期情况,同时丰富鱼类活体测定基因组大小的检测材料。以麦穗鱼(Pseudorasbora parva)为实验材料,采用流式细胞仪测定了7种不同组织的DNA含量并对其细胞周期进行了分析。结果表明:麦穗鱼的血液、肌肉、鳍条、鳃、肝脏、肾脏和性腺7种组织的DNA含量存在不同程度的差异,几种组织DNA含量大小排序依次为:肌肉>血液>鳍条>肝脏>性腺>肾脏=鳃,肌肉的DNA含量最高,达到3.20×10-6 μg,肾脏和鳃组织DNA含量最低,仅为3.05×10-6 μg,鳍条组织DNA含量与血液DNA含量相近,可以作为活体测定基因组大小的材料;血液和其他几种组织一样都表现出了一定的细胞周期现象,其中性腺和肝脏组织的G0/G1期时相的比例较低,并且这2种组织的细胞增殖指数比较高,分别为17.81%和13.19%,而鳃组织的细胞增殖指数最低,仅为6.54%,同时几种组织的细胞周期时相也存在着不同的差异。

Abstract:

In order to study the DNA content and the cell cycle of different tissues of fish, as well as enrich the material which was used to determine the genome size of fish in vivo, Pseudorasbora parva was used as experimental material, and flow cytometry was used to determine the DNA content and analyse the cell cycle in different tissues. The results showed that there were some differences in DNA contents in blood, muscle, fin, gill, liver, kidney and gonad of Pseudorasbora parva, and the highest and lowest tissue of DNA contents was the muscle and kidney, gill, which reached 3.20×10-6 μg and 3.05×10-6 μg, respectively. DNA contents in fin were similar to that in blood, so the fin could be as a material to determine the genome size of Pseudorasbora parva in vivo. All tissues of Pseudorasbora parva shown the cell cycle, the ratio of the time phase of G0/G1 in gonad and liver was the lowest, and the two tissues shown higher cell proliferation index that was 17.81% and 13.19%, respectively, while the gill was detected the lowest cell proliferation index which was merely 6.54%, there were also differences about the time phase of cell cycle in different tissues.
