基因枪法将细胞周期蛋白基因CycD2导入小麦的研究

摘要: 以小麦品种绵阳11作为基因枪转化的靶材料,用含有细胞周期蛋白基因CycD2和新霉素磷酸转移酶基因NPTII的质粒PBI121轰击小麦成熟胚性愈伤组织,在含有遗传霉素G418的筛选培养基对愈伤组织进行筛选后诱导得小麦再生植株,经PCR检测有4个转基因小麦植株呈阳性,Southern-blot鉴定后进一步证明CycD2基因已经被成功导入小麦并整合到小麦基因组中。

Abstract: The present study utilized mature seeds of wheat (Triticum aestivum L.) cultivar mianyang11 as the target material cultured in vitro, and the plasmid PBI121 which contains the cycd2 cell cycle protein gene and selectable marker NptII gene had been transferred into callus initiated from mature embryo of wheat by micro-projectile bombardment. The result showed that some transformation wheat could be refrained effectively and regenerated from the selection medium containing G418 solution. Furthermore, four plants were identified to be the transgenic ones containing the target genes of CycD2 by PCR and Southern-blot analysis.
