猕猴桃高效离体再生体系的研究

摘要: 为建立猕猴桃高效离体再生体系,试验以猕猴桃离体茎尖和茎段作外植体,在初代培养诱导试管苗成功的基础上,分别以MS和1/2 MS为基本培养基,附加不同的激素组合,研究不同培养基对猕猴桃叶片和叶柄芽诱导的影响和对猕猴桃生根的影响。试验发现:在MS+1.0 mg•L-1 6-BA+0.1 mg•L-1NAA的培养基中培养,初代苗诱导成功、生长良好;在MS+2.0 mg•L-1 6-BA+0.1 mg•L-1 NAA的培养基中,叶片和叶柄芽诱导率分别为137.50%和512.50%;在1/2 MS+0.9 mg•L-1 IBA和1/2 MS+ 0.6 mg•L-1 IBA的培养基诱导生根效果好,总根长,平均根长以及生根数均高于其他4组处理的结果;以消过毒的珍珠岩和草炭土(3∶7)为炼苗移栽基质,猕猴桃组培苗的成活率达到80%。试验结果为猕猴桃繁殖推广奠定了基础。

Abstract: To establish the high regeneration system of Kiwifruit in vitro culture the stem segment and apex explants in Actinidia was studied in this paper. After having gained in vitro plantlet successfully different of hormonal concentration combinations were added in MS and the 1/2 MS medium to study the effect of hormonal combinations with different concentrations on shoot induction and root induction. In initial culture of stem segment the plantlet had been gained successfully and grown well in the medium of MS +1.0 mg•L-1 6-BA +0.1 mg•L-1NAA. In the medium of MS +2.0 mg•L-1 6-BA +0.1 mg•L-1 NAA the rates of induction budding from leaf and petiole were 1.38, 5.13 respectively. The results showed that concentrations of IBA was 0.9 mg•L-1 and 0.6 mg•L-1 added in 1/2 MS in rooting culture medium were effective to shoot in vitro rooting, of which the total root length, the average root length and root number was higher than other treatments. The survive rate of plantlet transplanted into the substrate composed of 30% perlite and 70 % peat-moss was 80%.The result of this study has laid a foundation for rapid propagation in kiwifruit.
