芽孢杆菌BJ-6抗血清制备及其ELISA检测条件初探

Preparation of antiserum and development of an indirect ELISA to Bacillus BJ-6 strain

摘要: 芽孢杆菌BJ-6分离于杏树根部土壤,对多种植物病原真菌有抑菌活性,试验制备其抗血清,明确ELISA反应条件,用于相似抗原性生防菌的筛选。本文分别以芽孢杆菌BJ-6的抗菌蛋白和菌体研磨物为免疫抗原,制备出两种抗血清,其效价分别为1:3200和1:6400。间接ELISA反应发现:所购辣根过氧化物酶标记羊抗兔效价为1:7000;待测抗原可在37℃3h、4℃18h+37℃3h、37℃3h+4℃18h和4℃18h条件下包被;初步测定两种抗血清具有种的特异性。

Abstract: The strain of Bacillus sp. BJ-6 was isolated from the soil of apricot trees and showed strong inhibitory activity against plant pathogens. Two antisera of Bacillus sp. BJ-6 were successfully prepared by immunized the healthy male rabbit with the antifungal protein antigen and the somatic antigen. In the indirect ELISA, The titers of the two antisera were 1∶3200 and 1:6400. The titer of HRP-labeled secondary antibody was 1:7000. In addition, the two antisera showed species-specific to Bacillus amyloliquefaciens. Key words:Bacillus;antiserum;preparation;ELISA
