根癌农杆菌介导获得粳稻转基因植株

摘要: 以水稻粳稻品种空育131、垦鉴稻6号、垦鉴稻7号为材料,用根癌农杆菌感染成熟胚诱导的愈伤组织,将反义蜡质基因导入受体材料。经两次抗性筛选后,转入分化培养基中分化培养后获得转基因小苗。将转化植株进行GUS染色、PCR鉴定,能证明外源目的基因已经整合到植株中。

Abstract: The varieties are Kongyu131, Kenjiandao6 and Kenjiandao7 of Oryza stiva L subsup. Japonica. Agrobacterium tumefaciens infected maturate embryo of rice in order to induce callus. By this way, waxy gene can be transferred into accepted materials. After twice resistance selection, transgenetic seeding can be obtained in differentiation culture medium. Transgenetic plant is dyed by GUS and be identified by PCR, which can testify that exterior gene has been transferred into Japonica plant.
