犬细小病毒VP2基因序列分析

Sequence Analysis of Canine Parvovirus VP2 Gene

摘要: 2006~2007年间,从中国农业大学动物医院采集的犬细小病毒粪便样本中随机抽取20份,采用PCR技术,扩增了这20份样本的犬细小病毒VP2全基因序列。经核苷酸及推导的氨基酸序列分析发现,20个样本中,有19个样本属于CPV-2a,1个为CPV-2b,且所有病毒样本的297位均发生Ser→Ala的置换;基因型为CPV-2a病毒样本的555位均发生回归性置换(Ile→Val)。与经典的CPV-2a/b毒株比较,部分样本还出现了新位点氨基酸置换现象。其中,样本CPV/BJ005/07与CPV/BJ008/07 VP2的139位氨基酸残基由Val置换为Ile;80%(16/20)的样本在324位有Tyr→Ile的置换;样本CPV/BJ004/07和CPV/BJ050/07分别在226(Ser→Asn)与382(Arg→Lys)位发生置换。运用DNAStar软件对上述置换的氨基酸残基进行综合分析发现,除Ile-139突变意义不大外,Ile-324、Asn-226和Lys-382均处在VP2蛋白潜在的抗原表位区域,有着重要的生物学意义。

Abstract: Using polymerase chain reaction (PCR), we amplified the complete VP2 gene of twenty canine parvovirus samples collected in small animal hospital of China Agricultural University from 2006 to 2007. sequence analysis of nucleotide and deduced amino acid residues revealed that nineteen samples were CPV-2a and one CPV-2b among the twenty canine parvovirus. All samples had an amino acid substitution of residue 297 in VP2, Ser to Ala. nineteen CPV-2a samples studied in this study had reversion substitution (Ile→Val) at residue 555 in VP2. Compared with classical CPV-2a/b isolates, some samples had new amino acid substitutions: Samples CPV/BJ005/07 and CPV/BJ008/07 at amino acid residue 139 in VP2 was Ile instead of Val; Ile-324 variants accounted to 80% (16/20). Amino acid substitutions of residue 226 (Ser→Asn) and 382 (Arg→Lys)were found in samples CPV/BJ004/07and CPV/BJ050/07, respectively. All (Ile-139, Ile-324, Asn-226, and Lys-382) except Ile-139 substitutions were all in regions of potential antigen epitopes by DNAStar software analysis, and these substitutions may have an important influence to viral biology aspect.
