氟乐灵诱导西葫芦染色体加倍研究

摘 要:【研究目的】该试验利用氟乐灵对西葫芦染色体进行诱导加倍研究。【方法】方法1:浸种法 利用0.001%、0.005%、0.01%的氟乐灵溶液浸泡露白的西葫芦种子,浸种时间为4 h、6 h和8 h,通过根尖压片染色体计数法检测诱导率;方法2:生长点滴液法 以0.001%、0.003%、0.005%和0.01%的氟乐灵对不同品种西葫芦幼苗进行生长点滴液处理,每天17:00~18:00滴液一次,共滴6d,通过保卫细胞叶绿体计数法检测诱导率。【结果】结果显示:浸种法以浓度0.01%氟乐灵浸种6小时诱导效果最好,诱导率为25.6%;生长点滴液法以浓度0.005%的诱导效果最好,诱导率为20%;品种“京葫1号”、“长青1号”和杂交组合2-9×1604-1的四倍体诱导率分别为26.7%、20%和10%。【结论】氟乐灵可以诱导西葫芦染色体加倍,不同基因型西葫芦对氟乐灵的敏感性有差异。

Abstract: Abstract: [OBJECTIVE] The purpose of research is to induce pumpkin chromosome doubling with trifluralin.[METHOD] Method 1: the method of seed socking In the experiment germinative pumpkin seeds were socked with 0.001%,0.005% and 0.01% trifluralin solution for 4h,6h and 6h. The cell of root tip was inspected by microscope. Method 2: the method of dripping apical point With 0.001%,0.003%,0.005% and 0.01% trifluralin solution drip apical point seedling of different variety of pumpkin to induce chromosome doubling, the resut was detected with the number of chloroplast of leaf guard cells.[RESULT]The result showed that: about method 1 the best treatment for inducing tetraplont was obtained by socking seeds with 0.01% of trifluranlin solution for 6 hours, the induction frequency was 25.6%; about method 2 the result indicated that the best concentration for inducing tetraplont was the one of the 0.005% trifluralin solution, the induction frequency was 20 %.Among different variety, the induction frequency of tetraplont “Jing-hu No.1”,“Chang-qing No.1” and“2-9×1604-1”pumpkin seedling is respectively 26.7%,20% and 10%, [CONCLUSION] Trifluralin is able to induce pumpkin chromosome doubling and the sensitivity of different genotype of pumpkin to trifluralin is different.
