三聚氰胺及其衍生物对比格犬肾脏和睾丸的毒性评价

摘要:

试验将30只8月龄的健康雄性比格犬随机分成五组,对犬灌服含有不同剂量组合的三聚氰胺和三聚氰酸混合胶囊,以研究不同浓度的三聚氰胺及其衍生物对犬肾脏和睾丸功能的影响。在试验周期29天内,获得五组犬的死亡率、尿沉渣检出率和尿生化检测值(UPRO/UCRE), 并进行病理组织学检查。试验结果表明:①在给药浓度为30mg/kg体重三聚氰胺和10mg/kg体重三聚氰酸混合时,犬死亡率最高(66.7%);②检出尿沉渣种类有磷酸铵镁结晶、黄色结晶、针状结晶、肾小管上皮细胞、鳞状上皮细胞、移行上皮细胞,尿沉渣检出率随着摄入浓度增大;③各组犬的尿生化检测值(UPRO/UCRE)值基本在正常范围内;④病理组织切片中显示高剂量组犬肾中有结晶形成、间质性肾炎和脂肪细胞变性的等病理变化,但对睾丸组织无明显变化。由此可以得出结论:高浓度的三聚氰胺及其衍生物主要影响犬的肾功能,而对睾丸无明显毒害作用。

Abstract:

This study was aimed to investigate the effect of different dosages of melamine and cyanuric acid on the renal and orchic function of dogs. In these experiments, total 30 healthy male beagles (8-months old) were randomly divided into 5 groups, and they were fed with different dosages of capsules which had already been mixed by melamine and tricyanic acid. During the first 29 days of these tests, the mortalities of 5 groups, the composition and their ratioes of urinary sediment and the proportionalities of UPRO/UCRE were obtained. The results showed as following: ①the mortality of Group 4 was the highest one among all the groups (66.7 %); ②urinary sediment were composed by ammoniomagnesium phosphate crystal, yellow crystal, acicular crystal, epithelium of renal tubule, squamous cell, and transitional epithelium;the urinary sediment relevance ratio grows along with the dosage increase and the period extending; ③the proportionalities of UPRO/UCRE of each group were nearly normal on the whole;④Arrest of pathological changes, such as the form of crystallization, the nature nephritis and the fatty cell denatured occoured in higher dose of melamine and cyanuric acid in kidney but not in testis. Conclusion: it might be concluded that the high dosage of melamine and cyanuric acid be harmful to the renal normal function of male Beagles, but not influence it in their testis.
