固态发酵麸皮和玉米芯生产拟康氏木霉(Trichoderma pseudokoningii)孢子的实验研究

摘要:

本研究以麸皮和玉米芯为主要原料,筛选孢子产量最大的拟康氏木霉固态发酵培养基。采用单因子试验对麸皮、玉米芯比例,培养基含水量,其他佐剂进行了优化。浅盘式固态发酵法进行了模拟发酵。结果表明最优培养基:麸皮、玉米芯按8:2比例混合,调整含水量至55%~60%,添加0.1% KNO3,1%CaSO4,1%蔗糖。在28℃,接种量10%的条件下,利用浅盘式发酵进行拟康氏木霉孢子生产,1千克干料,发酵产出470g分生孢子及菌丝,每克发酵产物含1.34×109孢子。本研究为大规模工业化固态发酵生产拟康氏木霉孢子制剂提供了理论依据。

Abstract:

Using wheat bran and corncob as main material, the solid state fermentate medium of Trichoderma pseudokoningii which produce the most spores was screened. Single factor experiments were studied on the proportion of wheat bran and corncob, water content, and the additives. The optimized medium is composed of: wheat bran: corncob=8:2, water content 55~60%, KNO30.1%, calcium sulfate1%,sucrose1%. Shallow tray fermentation experiment was performed, the results show that under the conditions of 28℃, inoculum size of 10%, and the optimized medium, 470g dried mycelium and spores were obtained from 1000g dried raw material. One gram of dried fermentation products contains 1.34×109 spores. The research provides a theoretical basis for mass production of Trichoderma pseudokoningii mycelium and spores agents.
