花叶毛白杨叶片致黄因子的病毒学研究

Study on the Cause of Leaf Etiolation Virus of Populus tomentosa ‘Huaye’

摘要:

为了探索花叶毛白杨的花叶形成因子,以花叶毛白杨为研究材料,采用接种鉴定、ELISA测定法、病毒双链RNA检测及RT-PCR检测4种方法对花叶毛白杨的致黄因子进行病毒学鉴定。结果表明:(1)花叶毛白杨花叶样品过滤液经对指示植物烟草(Nicotiana tabacum)、大豆(Glycine max)及北京杨幼苗(Populus beijingensis)摩擦接种或注射接种后,未出现皱折、局部失绿等症状。(2)花叶毛白杨花叶样品过滤液选用24种抗血清进行酶联免疫检测,均未得到阳性结果。(3)花叶毛白杨花叶样品dsRNA电泳图未出现明亮条带。(4)RT-PCR未得到扩增产物。花叶毛白杨的叶片致黄因子不是由植物病毒引起。

Abstract:

In order to search factors related to the forming of mosaic leaf of Populus tomentosa ‘Huaye’which was used as the research material, Nicotiana tabacum, Glycine max and Populus beijingensis were used as show plants. Inoculability, ELISA, distill RNA test and RT-PCR method were used to detect yellow mosaic virus of Populus tomentosa ‘Huaye’ .The results showed that: (1) The plant of Nicotiana tabacum, Glycine max and Populus beijingensis didn’t show wrinkle and losing green after rubbing or injecting inoculability by using the percolate of Populus tomentosa ‘Huaye’. (2) ELISA didn’t show masculine. (3) The electrophoresis fig of Populus tomentosa ‘Huaye’ didn’t show bright strip. (4) There were no amplified products by RT-PCR. Plant Virus didn’t cause yellow mosaic of Populus tomentosa ‘Huaye’.
