丁香酚脂质体制备工艺研究

摘要:

目的:制备丁香酚脂质体并对其进行质量评价。方法:采用薄膜-超声法制备丁香酚脂质体;利用正交实验优化出最佳处方和最优制备工艺;以萃取法结合紫外分光光度法测定丁香酚脂质体包封率;并利用透射电镜观察其粒径形态及大小。结果:丁香酚脂质体最佳制备工艺和处方为:胆脂比1:6;药脂比1:5;超声时间40min;制的脂质体粒径均匀,呈球形小囊泡,包封率为(90±3)%。

Abstract:

Object: To prepare liposome of eugenol and evaluate its quality. Method: The orthogonal design was used to optimize the prescription and preparation technology of Eugenol liposome. The liposome of Eugenol was prepared with the film dispersion-supersonic method. The result of preparation was evaluated with the encapsulation rate which it was determined by UV spectrophotometry after extraction from Eugenol liposome. The appearances and the particle diameter of Eugenol liposome were examined with transmission electron microscopy. Results: The optimum prescription and preparation of Eugenol liposome were 1:6 (proportion of Cholesterin to Phosphatidylcholine), 1:5 (proportion of eugenol to Phosphatidylcholine), 40 min (ultrasonic time). The encapsulation rate of Eugenol liposome was 88.3%±3%. The particle diameter of Eugenol liposome was about 200nm and its appearance was uniformity globular vesicle.
