高活性纤维素酶的分离纯化

摘要:

为了纯化纤维素酶粗酶液,通过采用硫酸铵分级沉淀、Sephadex G-100凝胶柱层析、DEAF FF弱阴离子交换柱层析的方法,研究纤维素酶粗酶液分离纯化的方法。结果表明:采用硫酸铵分级沉淀、Sephadex G-100凝胶柱层析、DEAF FF弱阴离子交换柱层析的方法对内切葡聚糖苷酶组分的分离纯化有效。通过试验研究得到一种内切葡聚糖苷酶组分,其比活力提高了5.89倍,回收率为22%。

Abstract:

To study the separation and purification of cellulase from crude enzyme, the method of ammonium sulfate fractionation, Sephadex G-100 gel filtration and DEAF FF weak anion exchange column chromatography method were used. The results showed that: the usage of ammonium sulfate fractionation, Sephadex G-100 gel column chromatography, DEAF FF weak anion exchange column chromatography is a efficient method to purify CMCase. The specific activity of CMCase increase 5.89 times purifed in this way, while the recovery rate was 22%.
