冬虫夏草中多糖提取、纯化及抗氧化性能的研究

摘要:

为分离、纯化冬虫夏草多糖及其抗氧化性的研究,以冬虫夏草多糖提取量为考察指标,考察了时间、料液比、温度等单因素对冬虫夏草多糖提取量的影响。利用响应面分析进一步考察了时间-温度、温度-料液比、时间-料液比等因素的交互作用。对得到的冬虫夏草粗多糖利用Sepharose CL-6B羧甲基琼脂糖凝胶柱进行分离、纯化;并进行了体外抗氧化研究。通过冬虫夏草多糖提取率的二次多项数学模型解逆矩阵,得到冬虫夏草多糖最优提取条件:时间2 h,料液比1:34,温度100℃。纯化后多糖有较好的抗氧化性,可有效去除羟基自由基和超氧自由基的活性。

Abstract:

In order to separate and purify polysaccharides in Cordyceps sinensis and study its autoxidation. The extract effect factor of polysaccharides in Cordyceps sinensis such as extraction time, temperature, solid/liquid ratio, were investigated based on the content of polysaccharides. The interactions of time-temperature, temperature-solid/liquid ratio and time- solid / liquid ratio, etc were investigated by response surface methodology. The polysaccharide was purified by Sepharose CL-6B gel filtration. Meanwhile, antioxidation of purified polysaccharides was studied. The Design Expert software was employed to inversely resolve the created quadratic regression model of the extraction yields of polysaccharides. The optimums conditions were as follows: the extraction time was 2 h, solid/liquid ratio was 1:34 and temperature was 100℃. The purified polysaccharides in Cordyceps sinensis showed stronger antioxidants. It could remove scavenging DPPH free radical and superoxide radical were investigated respectively.
