猪鼻拭子样品采集及制备方法的标准化

The Standardization of the Collection and Treatment Methods for Swine Nasal Swab Sample

摘要: 为了黏膜样本采集所用材料及方法标准,并增强所检测样本数据的可比性与稳定性。以猪鼻拭子样品为研究对象,结合猪鼻腔的解剖结构以及所采集样品的特殊性,对猪鼻拭子的采集与处理方法进行探讨并标准化。结果表明:通过标化的方法对所采集的116份猪鼻拭子样本进行总蛋白浓度测定,发现所有鼻拭子样本的总蛋白均值为(6.92±0.41) mg/mL,变异系数为32.58%;对其中7头猪在不同时间采集鼻拭子测定总蛋白,结果同一头猪的检测数据变异系数除1头为13.95%外,其余均>10%。表明标化的猪鼻拭子采集及制备方法使检测结果具备较好的稳定性与科学性。

Abstract: The comparison and stability of detecting data were determined by the standard of materials and methods applied to collect mucosal samples. This study combined nasal cavity construction in pig and the specificity of the samples to investigate and standardize the collection and treatment methods of nasal swabs. The results showed that the total protein of 116 nasal swabs, collected by the methods standardized in this study, were detected, found that the total protein mean of all nasal swabs was (6.92±0.41) mg/mL and coefficient of variation was 32.58%. Besides, total protein content of the nasal swabs collected in five different times from 7 pigs were detected and the coefficient of variation of each pigs were below 10%, except one was 13.95%. The standard of the collection and treatment methods for nasal swabs made the detected results more stability and scientific.
