药用植物裸花紫珠的组织培养与快速繁殖研究

Tissue Culture and Rapid Propagation of Callicarpa nudiflora

摘要: 以种子为外植体,MS为基本培养基,通过比较不同植物生长调节剂种类和浓度配比、培养条件以及生根苗的移栽基质,建立裸花紫珠组织培养快速繁殖体系。结果表明:外植体表面消毒以75%酒精预处理10 s,再用0.1% HgCl2浸泡10 min,效果最好;种子在MS基本培养基上萌发;丛生芽继代增殖的最适温度为28℃,最适光照强度为1500 lx;培养基MS+ 6-BA 2.00 mg/L+ NAA 0.05 mg/L适宜继代增殖,30天的增殖系数为10.87;培养基MS+ NAA 0.50~0.75 mg/L适宜诱导生根,生根率100%;生根苗移栽于河沙、珍珠岩和表土(1:1:1)的混合基质中,成活率96%,高生长量最大。运用该组培快繁技术,可以实现裸花紫珠的工厂化育苗。

Abstract: Using seeds as explant, MS as basic culture medium, the techniques of tissue culture and rapid propagation of Callicarpa nudiflora were studied by comparing with types and concentrations of plant growth regulators, culture conditions, types of transplanting medium. The results showed that in the MS basic medium seed germination be made; the most suitable procedure for sterilization was to soak explants into 0.1% HgCl2 solution for 10 min after pretreatment with 75% alcohol for 10 s. Suitable temperature of buds subculture and proliferation was 28℃, the optimum light intensity was 1500 lx. MS+ 6-BA 2.00 mg/L+ NAA 0.05 mg/L be suitable for subculture and proliferation , the propagation coefficient was 10.87 every 30 days; the most optimum rooting medium was MS+ NAA 0.50-0.75 mg/L, and the rooting rate was 100%. The rooting seedling were effectively transplanted in river sand, perlite and topsoil (1:1:1), the highest survival rate was 96%, and the largest amount of growth. Using the technology of tissue culture and rapid propagation of Callicarpa nudiflora can realize the industrialized seeding.
