Cloning and Sequence Analysis of a UFGT Gene Fragment from maple cultivar ‘Autumn Blaze’

摘要: UFGT(类黄酮糖基转移酶)是植物花色苷生物合成途径的最后一个酶。以槭树‘秋天火焰’秋季变色期叶片总RNA为模板,设计简并引物,采用RT-PCR方法,扩增出一条长397bp的cDNA片段,序列分析结果表明,该cDNA片段与GenBank中登录的荔枝、柑橘柚等物种的氨基酸同源性在70%左右,可知克隆到的核苷酸片段为槭树叶片UFGT基因的cDNA片段,Genbank登录号为KC140229。

Abstract: UFGT(UDP glucose –flavonoid-3-o-glycosyltranferase)is the last key enzyme in the biosynthesis of anthocyanins. Total RNA was isolated from autumn leaves of Acer×freemanii ‘Autumn Blaze’. Using degenerate primer, a new cDNA fragment of 397 bp was amplified by RT-PCR. Sequence analysis demonstrated that the deduced amino acid sequence shared above 70% identity with Litchi chinensis, Citrus x paradisi, etc. These results suggested that the cDNA fragment cloned in this study was from UFGT gene of maple. The accession number in GenBank is KC140229.
