药百合种子萌发及染色体倍性检测

Studies on Seeds Germination and Ploidy levels of Lilium speciosum var. gloriosoides

摘要: 以浙江省广布种药百合为材料,采用不同激素浓度的培养基进行种子诱导及萌发实验,以不添加任何激素的MS培养基为对照,对药百合种子萌发率、诱导率、诱导芽数及生长势等进行研究,同时,对药百合种子诱导所形成试管苗进行染色体制片常规技术分析。结果表明,添加不同激素浓度培养基能显著促进药百合种子萌发,最佳培养基为MS+6-BA1.0 mg.L-1+NAA0.1 mg.L-1,萌发率为83.3%,诱导率为88.9%,平均诱导不定芽数达2.5个;未经剥皮处理的种子在30 d时仍无任何萌动;将诱导后的种子转至MS基本培养基中,仍表现出与转接前一致的生长状态;93%以上种子属于子叶出土型,仅接种在高浓度6-BA(1.0 mg.L-1)及低浓度NAA(0.1~0.2 mg.L-1)培养基上的种子有少量表现为种子留土型,占4%;种子萌发试管苗染色体倍性为2n=2x=24,未发生遗传变异。

Abstract: Lilium speciosum var. gloriosoides, which is widely distributing in Zhejiang Province and some other domestic areas.The seeds of induction and germination medium adding different concentration of hormones were studied, the MS basal medium without any hormones was used as control. The germination rate, shoot induction rate,,number of shoots inducted were measured growth of plantlets were observed during the experiment Besides, the seeds gernated plantlets were analyzed using the conventional chromosome preparation in this paper. The results showed that medium adding hormones could enhance the germination rate significantly, the optimum medium was MS media supplemented with 6-BA 1.0 mg.L-1 and NAA 0.1 mg.L-1,the germination percentage, induction rate and average indefinite buds were 88.3%,88.9% and 2.5, respectively. Seeds without peeling treatment did not germinate after inoculating 30 days at all. The seedlings growing 60 days were transferred to MS medium, the growth condition sustained as before. More than 93% seeds belonged to epigeal germination style, only some seeds inoculated only in high concentrations 6-BA(about 1.0 mg.L-1) and low concentration NAA(about 0.1~0.2 mg.L-1) displayed a hypogeal germination styles. The ploidy levels of seeds induction plantlets were diploid(2n=2x=24), and no genetic variation was found.
