三步法分离纯化副干酪乳杆菌细菌素的初步研究

A Preliminary Study of“The Three-step Method”in Purification of a Bacteriocin from Lactobacillus paracasei

摘要: 为了提高副干酪乳杆菌(Lactobacillus paracasei)HD wy-1细菌素的分离纯化得率和活性。本研究主要利用三步法从副干酪乳杆菌HD wy-1发酵液中分离纯化得到抑菌物质副干酪乳杆菌细菌素,分别采用硫酸铵沉淀、CM Sepharose Fast Flow阳离子交换柱层析和Sephadex G-75凝胶层析分离技术对副干酪乳杆菌HD wy-1产生的细菌素进行纯化。结果表明:细菌素得率为39.93%,比活为1.56×103 AU/mg,是纯化前的2.87倍,得到的细菌素样品经Tricine-SDS-PAGE电泳出现清晰条带,大小约为72 KDa。通过该方法可以较好的对副干酪乳杆菌的细菌素进行回收。该工作为研究菌体内细菌素的理化性质及其开发应用提供了理论基础。

Abstract: In order to improve the yield and activity of bacteriocin produced by Lactobacillus paracasei HD wy-1, the purification of antibacterial substances was studied with the three-step method. The antibacterial substances were extracted and purified by the Ammonium sulfate precipitation, CM Sepharose Fast Flow ion-exchange chromatography and Sephadex G-75 gel filtration. It is shown that the final yield of bacteriocin was 39.93% and the specific activity was 1.56×103 AU/mg. The specific activity was increased 2.87 fold. Tricine-SDS-PAGE showed that its molecular weight was about 72 KDa. The method had shown good results, which provided theoretical foundation for physicochemical property and development of bacteriocin.
