散穗高粱与白壳苏丹草杂交F1及其染色体加倍植株ISSR分析

ISSR Analysis on Sorghum-White Hull Sudangrass of Cross F1 and Its Chromosome Doubling Plants

摘要: 为了明确散穗高粱与白壳苏丹草杂交F1及其染色体加倍植株遗传差异性及其遗传距离,为下一步新品种育成利用奠定基础和提供科学依据。试验利用ISSR标记技术对散穗高粱与白壳苏丹草杂交F1及其染色体加倍植株进行多态性位点比较分析。结果表明:6条适宜引物共扩增得到重复性好的清晰条带164条,其中144条多态性条带,多态性比率达到86.9%;材料间的遗传距离变幅为0.2690~0.5923,散穗高粱与散穗高粱×白壳苏丹草F1加倍植株之间的遗传距离最远;供试材料以遗传距离0.26为基准可分为散穗高粱和白壳苏丹草;散穗高粱×白壳苏丹草杂种F1;散穗高粱×白壳苏丹草杂种F1加倍植株这三类。

Abstract: The paper aims to clarify genetic differences and genetic distance of sorghum-white hull sudangrass of cross F1 and chromosome doubling plant of sorghum-white hull sudangrass hybrids F1 and lay the foundation and provide a scientific basis for further breeding of new varieties. ISSR markers for sorghum- white hull sudangrass of cross F1 and chromosome doubling plant of sorghum-white hull sudangrass of cross F1 were used to carry out the comparative analysis of polymorphic loci. The results showed that, ISSR markers were amplified with 6 suitable primers and 164 bands were obtained, among which the bands of polymorphic were 144, accounted for 86.9%. The genetic distance was ranged from 0.2690 to 0.5923, and the genetic distance between sorghum and doubling plant of sorghum-white hull sudangrass of cross F1 was further. Using the genetic distance value of 0.26 as threshold, the test materials could be divided into 3 kinds: sorghum and white hull sudangrass, sorghum- white hull sudangrass of cross F1 and doubling plant of sorghum- white hull sudangrass of cross F1.
