辽宁杨叶片高频植株再生体系的建立

摘要: 为了提高辽宁杨的抗逆性及扩大它的适生区域,本研究以辽宁杨叶片为外植体材料,通过对附加不同浓度BA和NAA的MS培养基的筛选,建立了辽宁杨叶片高频植株再生体系,其不定芽诱导率及生根率都达到100%,辽宁杨最佳不定芽分化培养基配方为MS+BA 0.3 mg/L+NAA 0.05 mg/L,生根培养基配方为1/2MS+IBA 0.3 mg/L,为辽宁杨的抗性改良及分子育种实践奠定了基础。

Abstract: In order to enhance its resistance to cold, disease and pests and enlarge its growing area, a highly efficient plant regeneration system with 100% shoot regeneration and rooting rate from leaf explants of P.×Liaoningensis in vitro was established by comparing different concentrations of BA and NAA on MS medium, and it was concluded that the best medium for shoot regeneration was MS BA 0.3 mg/L NAA 0.05 mg/L, and the best medium for rooting was 1/2MS IBA 0.3 mg/L. The results of this study will contribute to the improvement of the resistance of poplar and the study of the molecular breeding.
