槐猪和杜洛克猪瘦素及其受体基因的差异表达研究

Expression Comparison of Leptin Gene and Leptin Receptor Gene in Huai Pig and Duroc Pig

摘要: 为了解7月龄、12月龄的槐猪和杜洛克猪5个不同部位脂肪组织LEP和LEPR基因的表达差异。采用荧光定量PCR技术,使用2-△△Ct方法计算LEP和LEPR基因的相对表达量,结果表明:LEP基因在背部皮下脂肪下层的表达量最高,其次是背部皮下脂肪上层、腹部皮下脂肪和板油,在冠状动脉脂肪中表达最低;7月龄槐猪脂肪组织LEP基因的表达量与12月龄槐猪、7月龄杜洛克猪差异显著(P<0.05),12月龄槐猪脂肪组织LEP基因表达量与7月龄杜洛克猪差异极显著(P<0.01)。7月龄槐猪脂肪组织LEPR基因的表达量与12月龄槐猪相比,只有冠状动脉脂肪差异显著(P<0.05)。

Abstract: The study aims to find the differential expression of leptin gene (LEP) and leptin receptor gene (LEPR) in five adipose tissues of 7-month-old and 12-month-old Huai pig and Duroc pig. LEP gene and LEPR gene were amplified by fluorescence quantitative PCR, then relative expression was calculated by transferring CT value into 2- △ △ CT. Results showed that the expression of LEP was the highest in lower subcutaneous fat, followed by upper subcutaneous fat, abdominal subcutaneous fat and leaf fat, and was the lowest in coronary fat. Expression of LEP in adipose tissues of 7-month-old Huai pig had significant difference compared with that of 12-month-old Huai pig and 7-month-old Duroc pig (P<0.05). Compared with 7- month-old Duroc pig, expression of LEP gene in adipose tissues of 12-month-old Huai pig was extremely and significantly different (P<0.01). Compared with 12-month-old Huai pig, only the expression of LEPR gene in coronary fat of 7-month-old Huai pig was significantly different (P<0.05).
